B cells regulate luminal bile acid composition in the small intestine

چکیده
Abstract Recent studies have demonstrated that bile acids (BA) influence immune cell homeostasis in the gut. However, whether and how, host response can BA biochemistry is unknown. This an important question to answer because small shifts composition gut significant pathophysiological consequences. Previously, we CD19 −/−mice, which a defect B activation consequently aberrant humoral response, develop intestine (SI) enteropathy associated with abnormal luminal composition. −/−mice are not B-cell-deficient present multiple phenotypes. Here, used cell-deficient JHminus;/minus; mice more effectively define this interaction. Results of our experiments provide three key observations. First, J H−/−mice altered SI pools compared their WT littermates. Second, adoptive transfer cells into neonatal led development normal rescued from enteropathy. Third, incapable secreting IgA failed restore acid only moderately protected animals developing Collectively, results clearly demonstrate regulate synthesis mucosal may be particularly for driving effect.

Regulation of antibacterial defense in the small intestine by the nuclear bile acid receptor.
Obstruction of bile flow results in bacterial proliferation and mucosal injury in the small intestine that can lead to the translocation of bacteria across the epithelial barrier and systemic infection. These adverse effects of biliary obstruction can be inhibited by administration of bile acids. Here we show that the farnesoid X receptor (FXR), a nuclear receptor for bile acids, induces genes ...

We previously reported that alcohol drinkers with and without cirrhosis showed a significant increase in fecal bile acid secretion compared to nondrinkers. We hypothesized this may be due to activation by alcohol of hepatic cyclic adenosine monophosphate responsive element-binding protein 3-like protein 3 (CREBH), which induces cholesterol 7α-hydroxylase (Cyp7a1). Alternatively, the gut microbi...
متن کاملSystemic factors are trophic in bypassed rat small intestine in the absence of luminal contents.
Mucosal histology, crypt cell proliferation and brush border enzymes were measured in rats with varying degrees of jejunoileal bypass, in order to compare the effect of systemic and luminal factors on adaptive growth and differentiation (brush border enzymes) in small intestinal epithelium. Eighty five percent jejunoileal bypass caused a functional short gut; in intestine remaining in continuit...
متن کاملBile acid-FXRα pathways regulate male sexual maturation in mice
The bile acid receptor Farnesol-X-Receptor alpha (FRXα) is a member of the nuclear receptor superfamily. FRXα is expressed in the interstitial compartment of the adult testes, which contain the Leydig cells. In adult, short term treatment (12 hours) with FRXα agonist inhibits the expression of steroidogenic genes via the induction of the Small heterodimer partner (SHP). However the consequences...
